Current studies show that Peyronie's disease (plastic induration of the penis) is an inflammatory condition of the penis where the elastic layer (tunic albuginea) in the central tissue (corpora cavernosa) suffers an injury. Consequently, the affected area loses elasticity and when filled with blood does not stretch, instead causing the penis to curve in the direction of the scar. Peyronie's disease is the most common cause of abnormal penile curvature.

The process starts with the oxidation of various fats and protein molecules leading to inflammation of the elastic layer (tunic albuginea) and the cavernous body. This continues until collagen fibers are replaced with inelastic fibers forming characteristic fibrous plaques (hardened nodules) of this disease. These plaques can be felt under the skin, generally at the top or sometimes at the sides of the penis. Most of them dissolve naturally, but sometimes persist, causing issues such as:

• Abnormal penile curvature (often upwards)
• Erectile dysfunction and/or pain during erection
• Shortening, loss of thickness, and/or penile malformation (indentations, constrictions, hourglass shape)
